6. Core Job Responsibilities
A. Hands-on Enzyme Immobilization Execution
Carrier Matrix Preparation & Screening: Prepare and condition various solid supports (such as hydrophobic macroporous resins, acrylic beads, silica matrices, and magnetic nanoparticles) for immobilization runs.
Execution of Binding Protocols: Conduct diverse immobilization techniques including physical adsorption, ionic binding, entrapment (e.g., alginate beads), and covalent attachment using cross-linking agents like glutaraldehyde.
CLEA Formulation: Support the formulation and preparation of Cross-Linked Enzyme Aggregates (CLEAs) and carrier-free immobilized preparations to maximize enzyme loading.
Mass Balance Calculation: Calculate and optimize immobilization parameters, specifically evaluating Immobilization Yield (the percentage of enzyme bound to the support) and Expressed Activity (how active the enzyme remains after being bound).
B. Stability, Mechanics & Kinetic Evaluation
Recyclability Trials: Run multi-cycle batch reactions to determine the
4G+ 40 structural and mechanical reusability of the immobilized biocatalyst, tracking operational half-life and activity decay over successive rounds.
Leaching Analysis: Monitor and analyze reaction supernatants using protein assays (e.g., Bradford or BCA assays) to ensure the enzyme is tightly bound and not leaching off the support matrix into the product stream.
Altered Kinetics Testing: Run time-course sampling to generate raw data for apparent kinetics modeling (Kapp_m, Vapp_max), specifically testing for mass transfer limitations and diffusion constraints under different stirring speeds or flow rates.
C. Route Design Verification & High-Throughput Screening Biotransformation Assays: Set up and monitor parallel, microscale enzymatic reactions (using automated liquid handlers or deep-well plates) to screen immobilized enzyme libraries against target chemical intermediates.
